A multi-omics study of the medicinal and edible plant Amomum tsao-ko identified 183 alkaloids in fresh and dried fruits collected from three plant accessions. Researchers characterized the compounds with ultra-performance liquid chromatography coupled with tandem mass spectrometry and found that both genetic background and post-harvest drying significantly shaped the alkaloid profile.

The work focused on the alkaloid metabolome of A. tsao-ko, a plant valued for its distinctive flavor and health-promoting properties. Although its volatile oil components have been extensively studied, the diversity and functional potential of its alkaloid constituents had remained largely unexplored. The new study provides what the authors describe as the first comprehensive alkaloid profile of the plant.

Of the 183 alkaloids identified, 76 differed among the analyzed samples. The researchers subjected these differential compounds to network pharmacology analysis, which mapped them to 455 predicted protein targets. The analysis highlighted akuammigine and tryptamine as bioactive compounds that may converge on the PI3K-Akt signaling pathway, described in the study as a central regulator of metabolism and cell survival.

p>The pathway result is a computational prediction rather than direct evidence of signaling activity. The study did not show that the compounds activate PI3K-Akt in people or establish clinical benefits from eating A. tsao-ko. Instead, the network analysis identifies relationships that can guide subsequent investigation of the plant’s alkaloids and their possible multi-target biological relevance.

p>Molecular docking produced a second set of computational predictions. Selected alkaloids showed strong predicted binding affinities for core targets including epidermal growth factor receptor, AKT1, and PIK3CA. These results nominate the proteins for further testing but do not by themselves demonstrate that the alkaloids bind them under biological conditions or produce therapeutic effects.

p>The researchers also combined the chemical analysis with transcriptomic data. This integrative analysis linked the accumulation of akuammigine to differential expression of strictosidine synthase, abbreviated STR1. STR1 was identified as a candidate gene associated with akuammigine biosynthesis, but its functional role requires validation in additional experiments.

p>Together, the results connect measured variation in alkaloid abundance with candidate biosynthetic and pharmacological mechanisms. Metabolomics established the range of compounds present and the effects of accession and drying, while network pharmacology and molecular docking identified potential protein targets. The transcriptomic comparison added a possible gene-level explanation for one metabolite pattern, although the work did not directly confirm the proposed biosynthetic function.

p>The study suggests that drying measurably changes the alkaloid composition of A. tsao-ko fruit and that differences among accessions also influence the profile. Because the comparison included three accessions and two fruit states, the results establish patterns within the analyzed material rather than their prevalence across other populations, growing conditions, or processing methods. The findings offer a foundation for functional validation and further development of the plant as a value-added functional-food resource, without demonstrating human health benefits.
