A team of researchers has resolved the architecture of the Norrin signaling complex and used the structure to engineer proteins that reproduce features of the natural signal. The work reveals that two curved Norrin homodimers connect two FZD4–tetraspanin 12 heterodimers, while the low-density lipoprotein receptor-related protein 5 LRP5LA3 domain binds tetraspanin 12 and both Norrin dimers. This arrangement brings the intracellular tails of FZD4 and LRP5 into a defined proximity, providing a structural explanation for how the complex can activate signaling.

Norrin is a ligand for Frizzled 4, or FZD4, a cell-surface receptor that works with tetraspanin 12 and LRP5 to regulate blood-vessel development and the integrity of the blood–brain and inner blood–retina barriers. Disrupted Norrin signaling has been associated with retinal disease, blood–retina barrier defects and deafness. The study, reported in Nature Structural & Molecular Biology, focuses on the molecular mechanism by which the signaling complex forms rather than on treatment outcomes in patients.

Determining the structure was challenging because the assembly depends on several interacting proteins. The researchers used a de novo designed FZD4 cysteine-rich-domain binder as a molecular “glue,” allowing them to capture and determine the structure of a Norrin–FZD4–TSPAN12–LRP5 complex. The structure shows that Norrin acts as a curved dimer that bridges receptor-containing units. It is not simply a solitary ligand contacting FZD4, but part of a larger assembly organized through multivalent interactions.

LRP5LA3 occupies a central coordinating position in the resolved complex. It interacts simultaneously with TSPAN12 and the two Norrin homodimers, helping position the cytosolic FZD4 and LRP5 tails for signal activation. The findings therefore connect the extracellular geometry of the complex with the intracellular signaling machinery. They also establish a physical model for how binding among Norrin, FZD4, TSPAN12 and LRP5 can bring receptor tails together without requiring all contacts to occur within a single protein pair.

Building on the structure, the researchers designed bispecific binders combining the FZD4 cysteine-rich domain with the LRP5LA3 domain. These engineered proteins function as Norrin surrogates in signaling assays, and their potency increased when the surrogate molecules oligomerized. The result indicates that multivalent assembly is not only a feature of the natural complex but can also improve the activity of engineered mimics. The study provides a structure-guided route for developing Norrin or Wnt mimetics, although the reported evidence comes from molecular structural analysis and signaling assays rather than demonstration of clinical benefit.
