Researchers have developed a modified version of L-Pantolactone dehydrogenase (LPLDH) that significantly enhances the efficiency of whole-cell biocatalysis for producing D-pantolactone. Published in 3 Biotech, the study details a combined strategy of rational design and directed evolution to overcome the limitations of the wild-type enzyme from Amycolatopsis methanolica. The primary objective was to address the low catalytic activity and poor industrial applicability observed in the natural enzyme, aiming to create a more robust biocatalyst for the green synthesis of chiral pharmaceutical intermediates.

The team, led by Shichang Liu and colleagues, utilized AlphaFold2-predicted structures, molecular docking, and bioinformatic analysis to identify critical functional determinants within the enzyme. Through this computational approach, residue T271 was pinpointed as a key site for modification. The researchers constructed various variants to test the impact of specific mutations, focusing on how changes at this residue might influence the enzyme’s performance in a whole-cell system.

Among the constructed variants, the T271D mutant demonstrated a substantial improvement in whole-cell catalytic activity. Specifically, this variant exhibited an 8.2-fold higher conversion rate compared to the wild-type enzyme. This significant increase in efficiency suggests that the T271D mutation plays a crucial role in enhancing the enzyme's function. The finding provides a concrete example of how rational design can yield biocatalysts with superior performance for industrial applications.

To understand the structural basis of this enhanced activity, the researchers conducted 50-nanosecond molecular dynamics simulations. These simulations suggested that the T271D mutation may rigidify the surface loop comprising residues 163 through 177. Further analysis using circular dichroism (CD) spectroscopy showed that the overall secondary structure of the enzyme was largely preserved. However, local secondary-structure changes were observed, which were consistent with the hypothesis of loop rigidification proposed by the simulations.

These findings are consistent with, but do not directly prove, a loop-rigidification mechanism. The authors describe the T271D mutant as a promising variant for further development, providing a basis for the practical application of AmeLPLDH. This work contributes to the broader field of biocatalysis by offering an improved tool for the sustainable production of D-pantolactone, a key intermediate in pharmaceutical manufacturing.
