A genome-wide survey of bread wheat has identified 58 members of the ADP-ribosylation factor (ARF) family and pinpointed one of them, TaARFB1b-A, as a contributor to the plant's defenses against Fusarium head blight. The open-access study, published in BMC Plant Biology, systematically cataloged the ARF gene family in hexaploid wheat and then tested the function of a small set of family members in response to the fungal pathogen Fusarium graminearum. ARFs are highly conserved GTP-binding proteins found across eukaryotes, where they regulate vesicle trafficking and cellular signal transduction.
Although these proteins have previously been linked to plant growth, development, and stress responses, their roles in wheat resistance to Fusarium head blight had remained largely unknown. The new analysis classified the 58 wheat ARF members into three subgroups: true ARFs, ARF-like proteins, and SARs. A scan of the promoter regions of the TaARF genes revealed abundant cis-regulatory elements associated with light response, growth and development, abiotic stress, and biotic stress.
One group stood out. A triplet of homologous genes, TaARFB1bs, comprising TaARFB1b-A, TaARFB1b-B, and TaARFB1b-D, was consistently up-regulated in response to both the fungus and the two molecular patterns. The functional tests produced the study's central result.
Using a virus-induced gene silencing system in the wheat cultivar Shimai 4185, the researchers simultaneously silenced the TaARFB1bs triplet. Silencing significantly reduced the plants' resistance to Fusarium head blight, indicating that these genes contribute to the cultivar's baseline defense against the disease. Conversely, when the team overexpressed TaARFB1b-A alone using a virus-mediated overexpression system in the same cultivar, resistance to the disease was significantly enhanced.
The paired results, loss of resistance upon silencing and gain of resistance upon overexpression, mark TaARFB1b-A as a candidate gene resource for breeding or biotechnological approaches aimed at strengthening wheat against FHB. The identification of the full TaARF family provides a framework for further dissection of how vesicle trafficking and signal transduction components shape disease responses in the crop. The authors present the work as a theoretical and genetic stepping stone toward understanding the family's biological functions in FHB resistance.
The study was published as open-access research on 23 October 2026 in BMC Plant Biology by Xihe Du, Dexian He, and colleagues. Because the findings rest on gene silencing and virus-mediated overexpression in a single cultivar, the reported effects apply to the experimental system used, and the authors frame TaARFB1b-A as a candidate gene for further study rather than a proven solution for field deployment.
