Forensic scientists and clinicians who work with child sexual abuse cases have long operated under a widely cited rule of thumb: biological evidence recovered from a victim’s body becomes difficult, if not impossible, to detect beyond roughly 72 hours after the incident. A new retrospective study from Türkiye challenges the practical weight of that assumption, showing that interpretable male DNA can be recovered from anogenital swabs well past the conventional three-day window—in one case as long as 110 hours after the alleged event—while clothing and crime-scene samples proved dramatically more productive than body swabs overall.

The research, led by Özlem Durma of the İslahiye Branch Directorate of the Ministry of Justice’s Forensic Medicine Institution in Gaziantep, together with Sıtkı Tıplamaz and Fatih Hitami Usluoğulları of the Department of Forensic Medicine at Marmara University’s School of Medicine in İstanbul, was published in the International Journal of Legal Medicine. Drawing on case files from a child advocacy center between 2019 and 2022, the team analyzed a substantial evidence inventory: 417 swab samples, of which 361 were anogenital and 56 were taken from body surfaces, alongside 68 clothing submissions and 22 crime-scene examinations, all originating from 136 acute child sexual abuse cases.

The analytical backbone of the study was short tandem repeat profiling, the workhorse of forensic genetics. Autosomal STR analysis examines repeating DNA sequences scattered across chromosomes inherited from both parents, generating a profile that is unique to an individual with the exception of identical twins. Because these loci are inherited in pairs, one from each parent, a victim’s own DNA profile typically dominates any mixture recovered from a body swab, since the vast majority of cells on a child’s skin and mucosa are the victim’s own. When perpetrator cells are present in vanishingly small quantities—the so-called low-template regime—the victim’s signal can mask the offender’s contribution entirely.

That is where Y-chromosome STR analysis becomes decisive. Y-STRs target repeat sequences on the male-specific Y chromosome, which female victims do not carry. In a mixed sample containing abundant female epithelial cells and trace amounts of male cells, Y-STR amplification effectively strips away the female background and amplifies only the male haplotype, the pattern of alleles passed down paternal lines. The authors defined DNA positivity in their study as the recovery of an interpretable male DNA profile—based on autosomal STR analysis and/or a reportable Y-STR haplotype—that could not be attributed to the victim. This dual-approach definition proved important: Y-STR testing enhanced detection precisely in those low-template or autosomal-negative cases where standard profiling would have returned nothing usable.

The headline numbers are striking in their asymmetry. DNA positivity was achieved in only 20 of 136 cases, or 14.7 percent, when relying on swab samples. Clothing submissions, by contrast, yielded interpretable male DNA in 54 of 68 cases—a 79.4 percent success rate. Crime-scene examinations performed even better, with 18 of 22 submissions, or 81.8 percent, producing usable male profiles. The message for evidence collection protocols is unambiguous: while body swabs are indispensable, the garments worn during or after the incident and the environment where it occurred are frequently the richest repositories of foreign biological material, and they should never be overlooked in favor of body-focused sampling alone.

Timing, as expected, mattered. The researchers categorized the post-incident time interval, or PIT, into three bands: Day 0, Days 1 through 3, and Days 4 through 10. Swab DNA positivity was observed across a range of 5 to 110 hours after the incident, with the latest positive detection coming from an internal anal swab collected dry—a detail that underscores both the persistence of semen-derived DNA in protected mucosal environments and the influence of swabbing technique on recovery. Detectability declined with increasing PIT, consistent with the known biology of sperm cell degradation and clearance, but the mere existence of positives beyond the 72-hour mark argues against rigid cutoffs in evidence collection policy.

The study also examined how victims’ post-incident behaviors affected recovery. Bathing was associated with descriptively lower swab DNA detectability, an intuitive result given that washing mechanically removes spermatozoa and epithelial cells from body surfaces and, to a lesser extent, from internal cavities. Interestingly, no statistically significant association was found between clothing change and DNA recovery. This may reflect the resilience of dried semen stains on fabric—prior research has shown that sperm DNA in laundered stains can survive remarkably well, protected within textile fibers from environmental nucleases and UV exposure. For forensic practice, the implication is that victims and caregivers should still be encouraged to preserve and submit the clothing worn at the time of the incident, even if it has been changed or washed.

Anatomical location within the anogenital region also shaped outcomes. External swabs—those taken from the vulva and perianal area—produced DNA positives more frequently than internal vaginal and anal swabs. The authors caution that this finding should be examined with an extended sample size before firm conclusions are drawn, but the pattern is biologically plausible. External surfaces are in direct contact with deposited semen and saliva during the assault, whereas internal deposition depends on the specific act; meanwhile, internal swabs contend with rapid physiological clearance mechanisms, including vaginal acidity and peristalsis, which degrade or expel foreign material quickly.

The persistence question has profound operational consequences. National guidelines in several countries have historically recommended that acute sexual assault examinations be performed within 72 hours, based largely on adult studies of sperm detection. In pediatric populations, however, delays in presentation are common: children may not disclose abuse immediately, caregivers may delay seeking care, or the child may be unable to communicate what happened. Studies of children referred for possible sexual abuse have repeatedly shown that most medical examinations reveal no acute physical findings, making laboratory evidence one of the few objective pillars available to investigators. Extending the evidence-recovery window, or at minimum discouraging the reflexive dismissal of late presentations, could therefore change outcomes in a meaningful fraction of cases.

The Turkish team’s data support a philosophy of comprehensive evidence collection regardless of PIT or post-incident actions. Because Y-STR analysis can rescue information from samples where autosomal testing fails, and because clothing and scene samples retain DNA at high rates independent of victim hygiene behaviors, the study argues that forensic examiners should collect the full evidence kit—including garments, scene materials, and both external and internal swabs—even when the child bathed, changed clothes, or presented days after the event. The cost of over-collection is modest; the cost of under-collection is permanent evidentiary loss.

Methodologically, the study is a retrospective file review, and the authors acknowledge its limitations. Molecular results were obtained from official forensic laboratory reports rather than from uniform experimental protocols, meaning swabbing techniques, laboratory workflows, and interpretation thresholds may have varied across cases. The sample of 136 cases, while respectable for this sensitive field, limits the statistical power to detect subtle associations—hence the careful hedging around the anatomical findings. Still, the raw recovery rates speak clearly, and they align with an international literature showing that sperm and seminal DNA can persist in the anogenital tract and on fabrics far longer than older clinical dogma suggested.

For child advocacy centers worldwide, where physicians, nurses, and forensic examiners coordinate care under emotionally and legally charged conditions, the study offers a data-driven reassurance: evidence is more durable than commonly believed, and modern molecular tools—particularly Y-STR profiling—can extract probative material from samples that would have been written off a generation ago. As the authors conclude, biological evidence, especially from anogenital samples, may persist beyond the commonly applied 72-hour window, and the decision to collect should rest on the circumstances of each case rather than on the clock alone.
