Study finds no link between pesticide exposure and autism-related social difficulties

A nationwide study of more than 3,300 mother–child pairs has found no significant association between prenatal exposure to organophosphate pesticides and autism-related traits in children. The findings, published online in JAMA Pediatrics on August 24, 2026, offer one of the largest and most diverse examinations to date of whether these widely used agricultural chemicals may influence social communication and related behaviors during childhood. Researchers emphasized, however, that the results do not establish that organophosphate pesticides are harmless. Instead, they suggest that exposure levels measured in this cohort were not a major driver of autism-related characteristics between the ages of approximately 5 and 7.

Organophosphate pesticides are a class of chemicals designed to disrupt the nervous systems of insects. Their primary biological action involves inhibition of acetylcholinesterase, an enzyme that normally breaks down the neurotransmitter acetylcholine after it transmits a signal. Excessive acetylcholine can lead to persistent nerve stimulation, and high-level exposure in humans can cause serious neurological symptoms. Earlier epidemiological research has associated prenatal or childhood exposure to some organophosphates with reduced IQ, impaired cognition, attention difficulties, and symptoms related to attention-deficit/hyperactivity disorder. Because the developing fetal brain undergoes rapid growth and organization, scientists have long questioned whether even lower-level exposure could influence social development or contribute to autism-related traits.

The new investigation was led by researchers at NYU Langone Health and involved data from the Environmental Influences on Child Health Outcomes, or ECHO, Program, a National Institutes of Health-supported research network. The analysis included 3,339 pairs of mothers and children recruited through 20 medical centers across the United States. The size and geographic reach of the cohort allowed investigators to account for a broad range of environmental, social, and biological variables that can affect child development. Such factors may include maternal characteristics, socioeconomic conditions, health history, chemical exposures, and other aspects of the prenatal environment. Large, multisite cohorts can also reduce the chance that results are driven by an unusual population or by the conditions of a single region.

To estimate exposure during pregnancy, researchers analyzed urine samples collected from mothers while they were pregnant. Rather than measuring the original pesticide molecules directly, the laboratory tests looked for metabolites, the short-lived compounds produced as the body breaks down organophosphates. These metabolites are commonly used as biomarkers because many organophosphate pesticides are rapidly transformed and eliminated from the body. The tests detected organophosphate-related metabolites in urine from 99 percent of the mothers, demonstrating how widespread low-level exposure was in the study population. Outside agricultural and occupational settings, people in the United States may encounter these chemicals primarily through food, particularly conventionally grown fruits and vegetables, although household, environmental, and community exposures can also vary.

The researchers then compared prenatal metabolite concentrations with children’s scores on the Social Responsiveness Scale, a 65-item questionnaire used to measure social communication difficulties and behaviors associated with autism. The scale evaluates traits across a continuum rather than simply classifying children as having or not having a diagnosis. Questions address behaviors such as difficulty interpreting other people’s emotions, reduced eye contact, challenges in reciprocal social interaction, repetitive patterns, and a strong preference for routines. This approach allowed the investigators to study subtle differences in behavior among children who had never received a formal autism diagnosis. After adjusting for relevant factors, the analysis found no statistically significant relationship between measured prenatal organophosphate exposure and the children’s Social Responsiveness Scale scores.

The finding is important because autism is a complex developmental condition shaped by many interacting influences. Genetic studies have identified numerous variants and biological pathways associated with autism-related traits, but genetics alone does not explain every difference in development. Environmental exposures remain an active area of research, particularly during pregnancy, when the nervous system is forming. Previous studies of organophosphate exposure and autism-related outcomes have produced inconsistent results. Some smaller investigations reported associations, while others found none. Differences in study design, exposure levels, timing of urine collection, population characteristics, and behavioral measurements may help explain why the evidence has not been uniform.

Haleigh Cavalier, the study’s lead author, said the results should not be interpreted as evidence that organophosphate pesticides are safe. The study observed exposure levels that were generally representative of low-level exposure in the broader population, rather than the much higher concentrations that may occur in agricultural workers, people living near heavily treated fields, or individuals experiencing accidental poisoning. The researchers also cautioned that the absence of a detectable association in this analysis does not rule out effects at higher exposure levels or among groups with heightened biological vulnerability. A chemical may have different consequences depending on dose, timing, genetic background, nutrition, co-exposures, and the capacity of the developing body to process it.

One possible explanation raised by the investigators is that exposure through food may occur alongside the nutritional benefits of eating fruits and vegetables. Produce supplies folate, antioxidants, fiber, and other nutrients that support healthy growth and neurological development. Those benefits could potentially offset some effects associated with low-level pesticide exposure, although the study was not designed to test that hypothesis directly. The researchers also noted several limitations. Organophosphate metabolites disappear from the body relatively quickly, meaning that a single urine measurement may capture only a brief period of exposure. Concentrations can fluctuate from day to day according to recent diet, metabolism, hydration, and the timing of pesticide contact. Even repeated measurements during pregnancy may therefore fail to represent a mother’s full cumulative exposure or identify the most biologically sensitive developmental windows.

The investigators say future work will examine other pesticide classes, microplastics, and additional environmental contaminants, while also exploring whether genetic factors modify children’s responses to chemical exposures. Such studies may require repeated biological sampling, more detailed information about diet and residential or occupational conditions, and long-term follow-up extending beyond early childhood. The current results provide reassurance that, at the relatively low exposure levels observed, organophosphates were not strongly associated with autism-related traits in this large cohort. At the same time, they reinforce the need for careful exposure monitoring and research that distinguishes between everyday background exposure and the substantially higher levels that may occur in agricultural or occupational environments.
