Stress resistance declines early in the replicative lives of budding yeast cells, before they reach the point of permanently ceasing division, according to a laboratory study in Biogerontology. Researchers tracked cells with different reproductive histories under nine stressful conditions and found that stress-induced mortality increased with replicative age. The pattern was observed from daughter cells through mother cells that had produced an average of up to eight buds, showing that diminished survival under stress can emerge well before the end of a cell’s reproductive lifespan.

The investigators adapted a flow-cytometry-based pulse-chase labeling method to distinguish yeast cells according to their replicative histories and measure their survival under harsh environmental conditions. Baker’s yeast divides asymmetrically, allowing mother and daughter cells to be separated experimentally; mother cells typically stop budding after about 15–25 events under optimal conditions. Because yeast suspensions consist mainly of young cells, this approach provided a way to examine whether cells at earlier stages of their reproductive lives already differed in stress resistance. The experiment therefore addressed an early phase of replicative aging in a yeast model, rather than measuring only the final cessation of cell division.

The age-related pattern was consistent across all nine stress conditions tested: older cells experienced higher stress-induced mortality than younger cells. This suggests that loss of stress resistance begins as a gradual, replicative-age-dependent process and can generate heterogeneity within a clonal population, even when its members share the same ancestry. The study does not establish what cellular changes produce this decline or whether the same pattern occurs in animals or humans. Its results concern survival under the particular stresses examined and should not be interpreted as a general measure of lifespan or as evidence of a clinical benefit.

Tests of three established genetic determinants of replicative lifespan showed that they were largely unable to account for the early decline in stress resistance. Depleting mitochondrial DNA altered baseline stress resistance under some conditions but generally preserved the age-dependent decline across the tested stresses. Deleting FOB1, which extends replicative lifespan under optimal conditions, did not markedly alter the decline under either heat shock or acetic acid stress. Deleting SIR2, which shortens replicative lifespan, produced a steeper age–mortality relationship under acetic acid stress but not under heat shock. Together, these findings indicate that the genetic determinants of maximum replicative lifespan examined in the study differ from those controlling the early loss of stress resistance, although SIR2 can affect the latter in a stress-dependent manner.
