For centuries, Osaka was known as the “Kitchen of the Realm,” a commercial crossroads where food from across Japan flowed into the city. Yet what ordinary people actually ate has remained far harder to reconstruct than the meals of wealthy households described in cookbooks or the animal bones discarded in urban refuse pits. A new study is now offering an unusually intimate view of premodern diets by examining a substance that survived directly on the teeth of ordinary people: ancient dental calculus, the hardened plaque that can preserve microscopic traces of food for hundreds of years.
Researchers from the Graduate University for Advanced Studies, SOKENDAI, Aomori Public University, Kyushu University and other institutions analyzed dental calculus from eight adult commoners buried at the Umeda cemetery, located north of present-day JR Osaka Station. The cemetery contains human remains from Osaka’s premodern urban population, providing a rare opportunity to investigate the lives of people who left few written records. By combining palaeoproteomics—the study of ancient proteins—with stable isotope analysis of bone collagen, the team reconstructed aspects of individual diets that conventional archaeological evidence cannot reveal.
Stable carbon and nitrogen isotope ratios provided a broad dietary framework. The results indicated that the individuals consumed a combination of terrestrial plant foods and marine resources, consistent with diets expected in a major coastal Japanese city. Stable isotope analysis is powerful because chemical signatures in bone collagen reflect long-term patterns of food consumption. However, the method generally identifies broad categories rather than precise ingredients. It can indicate whether a person relied heavily on marine or terrestrial foods, for example, but it normally cannot distinguish red sea bream from flatfish, or identify whether a person consumed a particular plant, mammal or processed food.
The proteins trapped in dental calculus supplied that missing resolution. As plaque accumulates and mineralizes, tiny particles and molecules from food can become physically enclosed within its dense structure. Some dietary proteins remain sufficiently intact to be extracted and identified using mass spectrometry, a technique that measures the molecular masses of peptides and compares them with reference protein databases. In this study, the researchers identified 38 dietary proteins representing 17 taxa, including nine fish taxa, terrestrial and marine mammals, and several plants. Rather than producing a generalized picture of “fish consumption,” the analysis revealed the specific biological sources that appeared in the diets of individual people.
The most striking result involved red sea bream, or Pagrus major. Proteins associated with the muscle tissue of this fish were detected in six of the eight individuals. Sea bream has long been associated with celebration, prestige and high-status cuisine in Japan, making its widespread detection in the Umeda sample especially significant. The findings suggest that a food historically treated as a luxury was not restricted to elite tables. At least some urban commoners had access to it, either through local markets, seasonal trade or the redistribution networks that made Osaka one of the most important food hubs in premodern Japan.
The researchers also identified proteins from flatfishes, including tonguefish in the Soleidae family. These fish remain closely associated with cuisine in the Kansai region and around the Seto Inland Sea, where they are valued for their delicate flesh and suitability for traditional preparations. Their presence in ancient dental calculus offers more than a list of forgotten meals: it suggests that regional food preferences have deep historical roots. The results provide molecular evidence that some ingredients central to modern Kansai culinary culture were already being consumed by Osaka residents centuries ago.
Even more revealing were the proteins from salmonids, cold-water fish that do not naturally inhabit Osaka Bay. Their detection indicates that some foods consumed by Osaka commoners had traveled considerable distances before reaching the city. During the premodern period, coastal shipping routes connected Osaka with the Japan Sea coast and northern regions, including Hokkaido. These maritime networks transported seafood, agricultural products and other commodities across the Japanese archipelago. The salmonid evidence demonstrates that such long-distance trade was not merely an elite privilege or an abstract economic system; it could place geographically distant marine products on the tables of ordinary city dwellers.
Dental calculus also preserved evidence of foods that written sources describe only indirectly. Proteins from non-human terrestrial mammals were found in the calculus of three individuals. Formal restrictions on meat consumption existed in premodern Japan, but historical records also describe the consumption of wild quadrupeds as “medicinal meat,” a practice that allowed animal products to be eaten while avoiding or reinterpreting religious and social taboos. The new evidence supports those accounts at the level of individual bodies. It shows that such practices were not simply literary claims or exceptional anecdotes but formed part of the lived food environment of at least some urban residents.
Another unexpected discovery involved proteins from cotton seeds. Cotton seeds are not normally eaten as a whole food because they contain compounds that make them unsuitable for ordinary consumption without processing. They can, however, be used to produce cottonseed oil. Cotton was widely cultivated around Osaka during the Edo period, and the protein evidence raises the possibility that cotton-derived oil products entered the diets of local commoners. The finding illustrates the unusual sensitivity of dental calculus analysis: it may detect not only recognizable ingredients but also processed commodities whose original biological sources would otherwise disappear from the archaeological record.
Together, the findings challenge the assumption that lower-status urban populations lived on a narrow range of locally available staples. The eight people examined at Umeda cemetery appear to have encountered a diverse food system that included marine fish, distant trade goods, wild mammal products and plant-derived commodities. Their diets were shaped by Osaka’s position within an extensive network of production, shipping and commerce. At the same time, the researchers stress that eight individuals represent only a tiny sample of the thousands of people buried at the cemetery and the hundreds of thousands who lived in premodern Osaka. The study is therefore not a complete portrait of commoner cuisine, but a demonstration of what can be recovered when archaeological science moves from broad population trends to individual molecular histories.
Future research could compare dental calculus from samurai, rural residents, merchants and people living in other major cities, including Edo and Kyoto. Such comparisons may reveal how status, geography, occupation and access to transportation influenced diet. They may also clarify how quickly foods moved through historical trade networks and whether regional culinary traditions emerged from local production or long-distance exchange. By analyzing proteins preserved in an otherwise overlooked biological material, scientists are turning ancient teeth into records of markets, mobility and taste. The study suggests that the history of food is written not only in cookbooks and refuse pits, but also in the microscopic residue left behind by meals consumed by people whose voices were never recorded.
