A new study published in Schizophrenia Research suggests that genetic risk for loneliness may help explain why some people are more likely to develop psychosis. Using data from the EU-GEI study, researchers investigated whether a person’s polygenic “liability” for feeling lonely could translate into clinical risk later in life.

The team focused on polygenic scores—statistical summaries that aggregate the effects of thousands of genetic variants associated with a trait. Here, the trait was loneliness, and the goal was to test whether higher genetic liability predicts greater susceptibility to psychosis.

Crucially, the study did not treat genetics as destiny. Instead, it examined a mechanism: adolescence might be the developmental window where loneliness becomes a conduit linking genetic predisposition to later mental health outcomes.

The researchers modeled adolescent loneliness as a mediator. In technical terms, they tested a pathway in which polygenic liability increases the probability of experiencing loneliness during early teen years, which in turn raises the risk of psychosis.

Results supported this mediation framework, indicating that the association between loneliness-related genetic liability and psychosis risk was partially explained by reports or measures of loneliness in adolescence. Put simply, genetics appeared to elevate the odds of loneliness first—then loneliness appeared to move risk forward.

This pattern matters for prevention strategies. If loneliness in adolescence is a meaningful intermediary, then interventions that reduce social isolation or improve connectedness could potentially buffer downstream psychiatric outcomes, even for individuals with higher genetic risk.

The analysis draws strength from the EU-GEI cohort design and the use of contemporary polygenic methods. While such findings do not prove causality in a single step, they offer a plausible, testable route for understanding how social experience can interact with biological susceptibility.

Overall, the work frames psychosis risk not only as a purely biological endpoint but as a developmental process shaped by both inherited factors and social environments—making loneliness a potential target for future public-health and clinical initiatives.
