Skin cancer, ranking as the fifth most common cancer worldwide, continues to present formidable challenges despite advances in immune checkpoint blockade and targeted treatments.</p>
<p>The metabolic landscape of skin tumors like melanoma and cutaneous squamous cell carcinoma is marked by profound metabolic reprogramming, allowing malignant cells to thrive under hypoxic and nutrient-deprived conditions. Tumor cells aggressively consume glucose and other key nutrients, not merely fueling their own growth but also depriving immune effector cells of resources crucial for their anti-tumor functions. This competitive metabolism leads to an accumulation of byproducts such as lactic acid, which further modulates immune cell behavior and fosters an immunosuppressive environment.</p>
<p>Immune cells demonstrate remarkable metabolic plasticity, with their function intimately linked to specific metabolic pathways. Glucose metabolism governs immune cell activation via central signaling nodes including mTOR and hypoxia-inducible factor 1-alpha (HIF-1α), while amino acid metabolism involving glutamine and arginine shapes immune responses through pathways like ATF4 and aryl hydrocarbon receptor (AhR).
